Determining the class and subclass of a monoclonal antibody is important when selecting the best purification method. IgA and IgM are often best purified by size (gel exclusion) or affinity columns, whereas IgG2a and IgG2b can be purified with Protein A columns at a pH of 7-8. IgG1 monoclonals bind best to Protein A at a slightly higher pH of 8-9.

Utilizing only two dipsticks, this 5-minute assay provides ELISA-like sensitivity without the time and effort of an ELISA. The assay can be run on both tissue culture supernatant fluid and on mouse ascites fluid. Protocols are provided for both sample types.
